Getting to Naples by car is easy. Just follow the A1 from Rome. However, I would avoid this at all costs. Try to arrive by train or other means, because actually driving in Naples is a headache you don't want.
Only drive in Naples is you have a death wish or are practicing for the demolition derby. Neapolitan motorists drive twice and fast and three times as aggressively as your average Italian, and while red lights seem to be considered optional in the rest of Italy, here they're ignored entirely.
No city in Europe (save perhaps Palermo or Athens) is more stressful, aggravating, confusing, and downright dangerous to drive in. If you want to explore the Amalfi Coast by rental car, pick it up the day you leave Naples (or, better yet, take the train to Sorrento and pick up your car there).
Do yourself a favor and park you car as soon as you arrive in Naples.
