The bus depot for Palermo is on Piazzetta Benedetto Cairoli, just off Corso dei Mille on the east side of the Palermo Centrale train station.
Most bus lines have offices a few blocks to the north along Via P. Basalmo.
Paermo is well connected by bus with the rest of Sicily, as well as a few long-distance lines into mainland Italy (though, frankly, a plane or train is going to make much more sense than a 10+-hour bus ride if you're coming from the mainland).
